Protein chemistry

MicroCoat relies on many years of experience in custom protein purification and protein chemistry service. Based on validated and standardized working procedures, the entire spectrum of methods from purification to modification and conjugation is available. If standard methods do not apply, modifications or alternative strategies are developed. All services can be offered from lab-scale to preparative-scale and include advanced analytics.

 

Protein purification

All standard methods for purification of antibodies, antigens or enzymes are well established. For demanding specifications, specific resins for affinity chromatography or immunoadsorption can be provided. Frequently used methods are:

  • gel filtration
  • ion exchange chromatography
  • protein A/G/L
  • affinity chromatography
  • positive-/negative immunoadsorption
  • cross-adsorptions

 

Protein modification

Enzymatic or chemical modification can be required to reduce undesired non-specific effects or to improve sensitivity.

  • Fab cleavage
  • F(ab‘)2 cleavage
  • blocking of functional groups
  • crosslinking for immunization

 

Protein conjugation

Standard procedures are established for the conjugation of monoclonal and polyclonal antibodies, Fab and F(ab‘)2 fragments, antigens, peptides to:

  • biotin (conjugation to different functional groups: -SH, -NH2, carbohydrates)
  • digoxigenin 
  • fluorochromes (e.g. FITC, rhodamine, Cy dyes, phycoerythrine)
  • enzymes (peroxidase, alkaline phosphatase, ß-galactosidase)           
  • carrier proteins for immunization

In addition, various methods for labelling of nucleic acids are established.
